Is it possible for someone to make the Subscribers column at http://groups.drupal.org/groups sortable? I'm interested in quickly seeing the largest groups.

not possible
because of how views is architected, this is not possible. the subscriber count is a separate query from the query that retrieves the groups (i.e. a handler function manages the display of this field) . i don't know of an acceptable workaround for this.
Top 25 Drupal groups ranked by # of members
I was curious and wanted to kill 5 minutes so...
In response to Group activity data analysis as well as the post Chris wrote above, I took a quick look at the groups and here's what I found via the the low tech and very old fashioned cut 'n' paste method. Feel free to correct any mistakes or accidental omissions.
Of course, the number of members a particular group has doesn't necessarily mean that group is any more or any less important than other groups. Groups are groups and all are important to the Drupal community.
