I'm having an issue with Acquia Slate 6.x-3.x-dev running on Drupal 6.16. I'm using Firefox 3.6.3 and Chrome 4.1.x.
All of my pages look correct, with the exception of the frontpage. I redownloaded the Acquia Slate source. I have unloaded and reloaded Fusion Core and Acquia Slate many times. I have flushed the caches (both Drupal and Firefox). I have changed AS configuration any number of ways to no effect.
I have also created a new page, called "Start". This page looks perfect. When I go into Site Information and set "Start" to be the frontpage, it loses its formatting (one column all the way down the page, no background images).
Any ideas?

Acquia Slate has different
Acquia Slate has different page.tpl.php files. One for the front and one for the rest of the pages. I think this is what you are referring to.
Do you have a URL or screenshots?
What do you mean by formatting?
Have you looked at http://acquiaslate.fusiondrupalthemes.com/
Does your site work like that? If so then it's working properly and you would have to modify it to get the front page to look differently.

I have the latest files downloaded, but I only see one page.tpl.php file. Am I missing something?page-front.tpl.php is there. But I don't see the frontpage regions available in the manage blocks admin page.

This raises another question
Once I get the frontpage-specific page.tpl.php installed, how to I configure the blocks for that page as opposed to the default?

I haven't worked with Fusion or Skinnr but deleting the page-front.tpl.php will give you the same page layout(like all other pages) on the front.
You will have to delete this file every time you update the theme.
If your looking to get the Front Page layout it's supposed to have I'm not sure right now. I will take a look when I get back later otherwise I'm sure someone here knows whats happening.
Preface_Sidebar region is
Preface_Sidebar region is missing from acquia_slate.info (and from the blocks admin area), but page-front.tpl.php seems to be expecting it.
