When I look for the "layout for _________" on the Manage Fields tab (or Display fields) of the content type there is nothing there.
Works for me fine in Drupal 7.....I've done two fresh installs....and I think I must be missing something simple.
Am I missing a module?
Is there some other way...via the Display Suite tab...to get at choosing a layout?
Been banging my head on this for days.....

Unfortunately, Display Suite for Drupal 6 does not currently support selectable layouts. Its layout is hardcoded.
External modules?
Same problem here. Sadly, it's too late to port the site to D7.
It says in the documentation that the D6 version needs extra downloaded modules in order to use layouts. This is great news, but since the documentation is written for Drupal 7 there are no links to such modules anywhere. Well, anywhere I searched anyway.
Help? Or should I ditch DS in favor of modifying node creation forms by hand? I shudder at the thought though...

Display Suite for Drupal 6 has NO layout support, other than the single three column layout which comes with Display Suite. If you can find somewhere which says it does, please let me know so I can get it changed. If, on the other hand, you cant see any settings at all, you might need the Node Displays module, http://drupal.org/project/nd.

There is no support for node creation forms at all, like there is in 7.x. If you want to change form layouts in 6.x you might like to look at http://drupal.org/project/form_builder and http://drupal.org/project/nodeformcols
