I spoke to a Phase II Dev at DrupalCon this year, and they said that every instance of OP they spin up, they use Pressflow. I'm currently setting up a site with OP, Pressflow and Varnish. I'll post results here when we go live. Because of how resource intensive some of the modules are, I think it makes total sense. Not sure if you're looking at using memcache as well, or how much authenticated traffic you have, but I would recommend that as well.
