Does anybody have experience creating route planning tools using any location/gmap/openlayer modules? Basically, my goal is to input an arbitrary amount of locations (with a fixed start/end point) and find the shortest route between all the points.

I have made another example http://googlemapsapi.tinemuller.dk/node/3 and tried to use it INSIDE Drupal as explained here http://groups.drupal.org/node/79804 so maybe you can do something like this too because I don't think such modules are made for Drupal yet.
GetDirections module
Check out http://drupal.org/project/getdirections - one of my colleagues just discovered this last week - I'd totally missed it until then. It only integrates with gmap & location module currently. I'm hoping to find time to submit a patch to integrate it with OpenLayers and other mapping modules in Drupal, though I'm not sure when I'll get around to it... The module maintainer seems really enthusiastic and open to patches, so I'd encourage you to get involved if there's some more developments you'd like to see there.

Used Route planning in a funny and creative way :-)
I have used the great module iframe http://drupal.org/project/iframe to make this http://googlemapsapi.tinemuller.dk/ - try the examples in the menu Cartrip - Drupalcon map - Drupalcon cartrip. I think it looks great and it's functioning and no one can see that an iframe is used - I think. :-)

CloudMade module
Just an FYI. I just started the CloudMade module to ingrate Drupal with CloudMade directly. I aim on making this avery lite layer to add CloudMade integration happen (not trying to replace the power of OpenLayers). But their API provides routing, so I will put that in somehow. This is jsut started and doesnt do anything yet.
